Fit the roofing sealant into a caulking gun so that the point of the sealant points outward and away from the trigger.
Carefully cut the tip of the roofing sealant at an approximately 45-degree angle a quarter of the way down the tip of the sealant's tube.
Lift the loose shingle and squeeze the trigger of the caulking gun to lay down a bead of sealant underneath. Be sure to also apply the sealant to any visible cracks in the wood of the roof.
Push the shingle back on top of the wet sealant and firmly press it into place.
Place a heavy brick or paving stone on top of the shingle and wait overnight before removing it.
